Our liver eliminates toxic compounds through a two-step enzymatic process, also known as Phase I detoxification and Phase II detoxification (or biotransformation) pathways. Specific reactions occur in these two pathways that assist the breakdown and neutralization of unwanted toxic chemical compounds into harmless substances:

Phase I Detoxification (biotransformation)- This is usually the first defense against unwanted chemical compounds. A series of enzymes, called cytochrome P450, (composed of a group of 50 to 100 enzymes) use specific reactions and nutrients to chemically change harmful fat-soluble toxins into even more toxic substances called "intermediary metabolites". Since the intermediary metabolites are often more toxic than the original fat-soluble toxins, they have to go through Phase II detoxification in order to become less toxic. If intermediary metabolites are not further reduced into less harmful substances by Phase II enzymes, intermediary metabolites accumulate and can cause various illnesses such as allergies, frequent colds, flus, etc. A decreased activity of Phase II enzymes or an increased production of Phase I enzymes (which does not allow Phase II enzymes to catch-up with toxin overload) can prevent the detoxification from intermediary metabolites. Specific nutrients needed for Phase I detoxification are: B vitamins (vitamin B2, vitamin B3, vitamin B6, vitamin B12), folic acid supplement and various flavonoids including rutin and quercetin.

Phase II Detoxification (biotransformation)- During Phase II detoxification, the attachment (or conjugation) of specific molecules onto the intermediary metabolite occurs. This is where the liver cells convert the highly toxic intermediary metabolites into non-toxic, water soluble molecules called excretory derivatives. This process is accomplished through conjugation, or attachment, of specific molecules to the intermediary metabolite after passing through six Phase II detoxification pathways. Since excretory derivatives are water soluble, they can be eliminated through the urine, bile or stool.

The 6 Phase II Detoxification Pathways:

First Detoxification Pathway, Sulfation- This is the detoxification pathway where toxins attach with sulfur-containing compounds. This is the main detoxification pathway that neutralizes the stress hormone cortisol, as well as some commonly prescribed pharmaceuticals, food additives, chemicals produced by intestinal bacteria and environmental toxins. Sources of sulfur include the sulfur-bearing amino acids such as methionine and cysteine.

Second Detoxification Pathway, Glucuronidation- In this detoxification pathway, glucuronic acid combines with toxins. This in turn helps to detoxify certain medications (such as aspirin), preservatives, food additives (such as benzoates), reproductive and adrenal hormones. The detoxification pathway of glucuronidation requires magnesium and glucuronic acid to function properly.

Third Detoxification Pathway, Glutathione Conjugation- The attachment of glutathione to toxins assists in the detoxification and elimination of fat soluble toxins, especially heavy metals including mercury and lead. Glutathione is a very important antioxidant and anti-caner agent in the body. The production of glutathione requires the presence of amino acids such as cysteine, glutamic acid and glycine. Nutrients that increase glutathione levels include vitamin C, alpha-lipoic acid and the amino acids glutamine and methionine.

Fourth Detoxification Pathway, Acetylation- In this detoxification pathway, acetyl co-A is attached to toxins to make the less harmful and easy to excrete. In order for this pathway to work at its optimal level, vitamin B5 (pantothenic acid) and vitamin C are needed.

Fifth Detoxification Pathway, Amino Acid Conjugation- The conjugation of toxins with amino acids occurs in this detoxification pathway. The amino acids commonly used in this pathway include glycine, taurine and glutamine. These amino acids help to excrete many toxic chemicals (know as xenobiotics) from the body. Amino acids are found in protein-rich foods if they are eaten in adequate amounts.

Sixth Detoxification Pathway, Methylation- Involves conjugating methyl groups to toxins. This detoxification pathway is used to detoxify many steroid hormones, including estrogen. The principal amino acid methionine drives this detoxification pathway. Methylation also requires the cofactors vitamin B12, folic acid and choline to function properly.
